Medicore’s standard Hybridoma Development Program takes approximately 4- 6 months to complete. Our high rate of success is due to a large part to frequent reporting and communications with the client throughout the program.
We have divided the program into the following four main phases:
(1) Immunization Phase An antibody response is generated in mice to your specific immunogen. This phase includes: - Primary Immunization of 5 mice (50-100 micrograms immunogen/mouse)
- Boosts (50-100 micrograms immunogen/mouse)
- Serum collection
- ELISA to determine serum titers

The sequences of events are as follows: Day 0: Preimmunization serum collection and primary injection Day 28: First boost Day 38: Serum collection and specific antibody titration Day 56: Second boost Day 66: Serum collection and specific antibody titration. The titer should be strong enough to proceed for fusion phase. Day 98: Fusion boost (2) Fusion Phase On day 102, spleen cells from the selected mouse are fused with a myeloma cell line to develop a hybridoma clones that secrete specific antibody. On day 116: Fusion screening is done. The fusion products are screened by ELISA. Catalog number: MAb-2
(3) Cloning Fusion products showing strong ELISA results are cultured and culture supernatant fluids are sent to the client for confirmation and selection of monoclonal antibody producing cultures. Further work is performed to subclone the parental screen. Finally, clones are selected and culture supernatant fluids are again sent to the clients for confirmation
