Ascites Production

Clones are grown and injected in mice for ascites production. Cells are injected to pristane primed mouse. Ascites production is the most cost effective method of generating antibody from hybridoma cell lines. We prime adult female Balb/c mice with pristane 7 - 14 days before injection of the cell line. The hybridoma cell line is injected into the peritoneal cavity at a concentration of 0.5 million cells per mouse. Ascites fluid generally develops within 10 to 14 days and harvested by gentle aspiration of the abdominal cavity. Each mouse is tapped up to 3 times. The amount of ascites production and the presence of specific antibody in ascites are variable.
